EU countries bin €4B worth of COVID vaccines
| At least 215 million doses of COVID-19 vaccines purchased by EU countries at the height of the pandemic have since been thrown away at an estimated cost to the taxpayer of €4 billion, an analysis by POLITICO reveals. And that's almost certainly an underestimate. |
Many of the vaccines in question were purchased at the height of the pandemic in 2021 when the EU, the U.S. and the U.K. were all scrambling to secure a limited number of doses. It was during that frenzied time that the EU entered into its single biggest contract to purchase 1.1 billion doses from Pfizer and BioNTech.
It's easy to forget how uncertain things were in 2021, and the EU deal was lauded at the time. But both the size and the timing of the agreement turned out to be problematic. Countries were locked into buying doses even as the pandemic subsided, while efforts to donate excess jabs to third countries were thwarted by falling demand and logistics issues.
The repercussions are more than financial. Questions around how the big Pfizer contract was negotiated have dogged Commission President Ursula von der Leyen ever since the New York Times reported that the EU chief had exchanged messages with Pfizer's CEO in the run-up to the deal.
The contract has already been renegotiated by the European Commission under pressure from EU countries suffering from a vaccine glut. Both Poland and Hungary stopped accepting vaccines and are being sued by Pfizer for non-payment. In Romania, prosecutors want to lift immunity for its former prime minister and two former health ministers, claiming excessive vaccine purchases caused more than €1 billion in damages to the state.
In the meantime, the jabs will keep coming, with the revised contract with Pfizer locking European countries into buying vaccines until at least 2027. |

Mucosal boosting enhances vaccine protection against SARS-CoV-2 in macaques
| It has been hypothesized that enhanced mucosal immunity will be required to block infection and onward transmission. Intranasal administration of current vaccines has proven inconsistent3-7, suggesting that alternative immunization strategies may be required. Here we show that intratracheal boosting with a bivalent Ad26 based SARS-CoV-2 vaccine results in substantial induction of mucosal humoral and cellular immunity and near complete protection against SARS-CoV-2 BQ.1.1 challenge. 40 previously immunized rhesus macaques were boosted with a bivalent Ad26 vaccine by the intramuscular, intranasal, and intratracheal routes or with a bivalent mRNA vaccine by the intranasal route. Ad26 boosting by the intratracheal route led to substantial expansion of mucosal neutralizing antibodies, IgG and IgA binding antibodies, and CD8+ and CD4+ T cell responses, which exceeded those induced by Ad26 boosting by the intramuscular and intranasal routes. Intratracheal Ad26 boosting also led to robust upregulation of cytokine, NK, T and B cell pathways in the lung. Following high-dose SARS-CoV-2 BQ.1.1 challenge, intratracheal Ad26 boosting provided near complete protection, whereas the other boosting strategies proved less effective. Protective efficacy correlated best with mucosal humoral and cellular immune responses. These data demonstrate that novel immunization strategies induce robust mucosal immunity, suggesting the feasibility of developing vaccines that block respiratory viral infections. |

Не думаю, что он позволил себе прямую ложь.
А по вашей ссылке разве общая смертность указана, а не зарегистрированная как смертность от ковида? Тегнелл говорит об общей избыточной смертности от всех причин. Я тут уже ссылался вот на эту табличку, оговариваясь, что её достоверность трудно проверить. Но вот и Тегнелл говорит то же самое. |

Я сильно не залюбил, когда у нас ввели комендантский час. Виноваты в этом, конечно, политики, а не врачи любого уровня (как и в провалах домов престарелых, как и в развале системы здравоохранения). Меня достаточно было попросить не выходить из дома, и я даже не требовал бы того же от других.
Тегнелл и "шведская модель" мне конечно нравятся, но надо понимать, что и там загрузка больниц регулировалась сортировкой, и более серьёзные меры не исключались из инструментария. |

На днях обнаружил, что пластиковые затяжки для жгутов уже больше не затягивают, хотя внешне на них зубчики есть. А для медицины еще более строгие требования. Ткань в масках синтетическая, может начать осыпаться, а мы эту труху вдыхаем. Да и степень фильтрации после осыпания ухудшается.

| Я, кстати, за все время медицинские N95 даже в руках не держал. Когда просто требовалось напялить, что изредка случалось, надевал обычную медицинскую маску. А когда для защиты, то респиратор с клапанами и P100 фильтрами. Такие не портятся, фильтры только можно менять. Обычные медицинские тоже, думаю, не портятся. |

Nearly 17,000 people may have died after taking hydroxycholoroquine during the first wave of COVID, according to a study by French researchers.

В общем на КД #здоровье зараженных мышей пострадало не так сильно, воспаление не разогналось, да и метаболизм аминокислот, нуклеотидов, липидов и клеточной энергии остались сохранными
Правда, насколько эффекты кетогенной диеты у мышей и людей сопоставимы — неизвестно. В общем, к дизайну есть вопросы, но мысль интересная.

Что произошло: сиквенс был подан в базу данных NCBI с не полностью заполненными административными полями формы, и потому оказался в системе «завешен», а автору отправлен запрос с уточнениями. Но поскольку в Китае к этому моменту уже началась беготня, которую все по телевизору видели, уточнения получены уже не были, и сиквенс свет не увидел |
Dr. Jesse Bloom, a virologist at the Fred Hutchinson Cancer Center, wrote Wednesday in an analysis of Ren’s submission that it “clearly falsifies the Chinese government’s claim that the causative agent of the Wuhan pneumonia outbreak still had not been identified near the end of the first week of January 2020.”
The earlier submission “would have provided adequate information to initiate vaccine production in late 2019 if it had been made public,” he said, noting that drugmaker Moderna “used the spike sequence to design its COVID-19 vaccine” within two days of the January 12 release...
“The belated discovery of the submission underscores the importance of rapid data sharing during outbreaks, since immediate public release of the sequence could have accelerated by several weeks the development of COVID-19 vaccines that saved thousands of lives per week in the United States alone,” he said.
Even two weeks “would have made a huge difference in the pandemic,” agreed Dr. Eric Topol, founder and director of the Scripps Research Translational Institute. The fact that the vaccine program began immediately on publication of the genetic sequence “shows you how important that sequence was.”
“When you sequence a virus – it’s not even just a vaccine – then you’ve nailed it. You know exactly the features, about the spike protein and all the other major components: the nucleocapsid, the envelope, the whole entire panoramic view of the virus. You can’t get that without the sequence.”
The documents should be read in the context of hindsight, says Dr. Kristian Andersen, an evolutionary biologist and director of infectious disease genomics at the Translational Institute.
“In late 2019, nobody knew that a pandemic would later ensue,” he wrote in an email. “This is a really critical part that most people seem to forget – nobody knew back then that a never-before-seen coronavirus only distantly related to SARS-CoV-1 was causing ‘mysterious’ illnesses in patients associated with a wet market in the middle of Wuhan, which would later spark a devastating pandemic.
“Should the sequence have been released at the time and [marked] as preliminary data? Sure, that would have been great, and is a good example of where we could hope to do better in the future,” he said. “Whoever reviewed the sequence at NCBI over the holiday period in 2019 would have no way of connecting this sequence to a ‘mysterious’ illness in Wuhan – because it was yet to be reported.” |

В квесте о происхождении COVID-19 важный прорыв.
Еще в 2022 году была опубликована теоретическая статья с анализом генома возбудителя ковида, РНК-вируса SARS-CoV-2. В статье говорилось, что этот вирус как будто собрали из шести примерно одинаковых частей, используя принятый в биотехе метод нарезки генома специальными энзимами. Участки сшивки этих частей типичны для синтезированных вирусов, но совершенно нетипичны для вирусов природного происхождения. Статью, конечно, пробовали осмеять, дескать, это "уровень детского сада".
Проматываем в 2024 год. Американская общественная группа "U.S. Right to Know" по запросу получила текст заявки на грант, направленный в 2018 г в DARPA (Агентство прорывных исследований). В заявке на проект под названием DEFUSE предлагалось создать синтетический высокозаразный для человека коронавирус из шести частей с использованием тех самых энзимов. И с фуриновым участком (furin cleavage site). Заявленная цель этого gain of function проекта - способствовать созданию эффективных вакцин. [А как же!].
Упомянутый участок расщепления фурина в геноме вируса отдельно давно вызывал вопросы, так как его нет ни в одном из 871 известных природных коронавирусов.
А кто подал заявку на грант? Питер Дащак (EcoHealth Alliance, США), Ральф Барик (Университет Северной Каролины, США) и Ши Чжэнли (директор Центра новых инфекционных заболеваний, Институт вирусологии Уханя).
Агентство DARPA финансировать проект DEFUSE отказалось. А поздней осенью 2019 года в Ухане началась пандемия коронавируса, геном которого состоит из шести частей и содержит фуриновый сайт.
Как пишет один биолог, новые данные переводят информацию о необычном строении генома SARS-CoV-2 из разряда "достойно внимания" в разряд "дымящийся пистолет на месте преступления".
Другие эксперты говорят, что пистолет, конечно, есть. Но не дымится. Проклятая неизвестность, короче.
